The attachment site for phage Phi80 is between the opp operon and the trp operon on the E. col chromosome. Aberrant excision of the Phi80 prophage integrated at this site can yield specialized transducing phage carrying either the opp genes or the trp genes. An example of how low (LFT) and high (HFT) frequency opp transducing lysates can be produced is shown below.
Note that because of constraints on the length of DNA packaged into the phage head, the more chromosomal DNA the phage carries, the less phage DNA can be packaged. The following example shows several potential specialized transducing phage that could be obtained by induction of a Phi80 lysogen. The length of phage and chromosomal DNA is indicated in Kb.
