The Guide to Phage Genomics:
Sequencing

OVERVIEW

Sequence random clones to get 8-10 fold coverage

There are many core facilities available throughout the US and UK for high through-put sequencing requiring various levels of work on the part of the interested investigator. If you know your genome size then you can calculate approximately how much sequencing will be required to get 8-10 fold coverage of the genome to estimate costs in evaluating your sequencing options.

Find good basic descriptions / tutorials of shotgun sequencing at: http://www.genome.washington.edu/UWGC/tutorial/shotgunhtm.htm and http://nema.cap.ed.ac.uk/teaching/genomics/Genomics3.html
